THEMIS enhances the magnitude of normal and neuroinflammatory type 1 immune responses by promoting TCR-independent signals

Signals that determine the differentiation of naïve CD4+ T helper (TH) cells into specific effector cell subsets are primarily stimulated by cytokines, but additional signals are required to adjust the magnitude of TH cell responses and set the balance between effective immunity and immunological tolerance. A publication in Science signaling (doi: 10.1126/scisignal.abl5343) by the group of Renaud Lesourne identified the T-cell signaling protein THEMIS as a selective amplifier of TH1 cell immune functions. This amplification mechanism promotes efficient immune responses against foreign antigens, but is detrimental in the context of central nervous system autoimmunity by increasing T-cell encephalitogenic responses against myelin-based antigens. This publication contributes to a better understanding of the fundamental mechanisms that control the magnitude of immune responses in normal and pathological contexts.
