Objectifs Scientifiques


La sclérose en plaques (SEP) est une maladie inflammatoire démyélinisante du système nerveux central (SNC) qui est la principale cause de handicap chez les adultes jeunes. En raison de sa prévalence croissante, son évolution chronique et l’effet partiel des thérapies actuelles, la SEP a des conséquences socio-économiques majeures.

L’objectif principal de notre équipe est de décrypter les mécanismes cellulaires et moléculaires des maladies inflammatoires qui affectent le SNC comme la SEP mais également d’autres maladies auto-immunes ou infectieuses du SNC.

Nos Projets


AXE 1: Les lymphocytes T dans les maladies inflammatoires du SNC: Migration, fonctions et regulations

Notre objectif majeur est de mieux comprendre les déterminants cellulaires et moléculaires responsables du vieillissement et des pathologies inflammatoires du système nerveux central (SNC).

 

Nos projets de recherche reposent sur l’utilisation de modèles expérimentaux uniques et d’échantillons biologiques de patients. Nous étudions la transmigration et les fonctions effectrices des lymphocytes T, en particulier des lymphocytes T CD8, en utilisant plusieurs modèles d’auto-immunité du SNC que nous avons récemment développés dans le laboratoire. Plus précisément :

1/ Nous étudions les mécanismes immunologiques sous tendant le développement de la narcolepsie de type 1, pathologie chronique du sommeil due à une élimination sélective d’une population de neurones hypothalamiques produisant l’orexine / hypocrétine. Dans ce but nous étudions les échantillons sanguins de patients atteints de narcolepsie de type 1, de sujets atteints d’autres affections du sommeil, et de sujets sains appariés pour l’âge, le sexe, et type HLA.  De plus, nous avons développé un modèle murin unique de narcolepsie auto-immune. Les mécanismes conduisant aux lésions de l’hypothalamus et le lien entre cette affection et la vaccination contre la grippe pandémique H1N1 de 2009 sont recherchés.

2/ Les Prof. Guillaume Martin-Blondel et Roland Liblau évaluent, à l’aide d’un modèle murin et dans des expériences de co-culture, les interactions entre lymphocytes T et cellules endothéliales de la barrière hématoencéphalique et leur rôle potentiel dans le développement d’affections inflammatoires du SNC (vieillissement, sclérose en plaques, syndrome de SUSAC, neuropaludisme …

3/ Dans le cadre des affections neurologiques paranéoplasiques nous évaluons le lien mécanistique entre une réponse immune anti-tumorale, les immunothérapies des cancers et le développement de l’auto-immunité dans le SNC

4/ En utilisant un modèle animal de sclérose en plaques, le modèle murin de l’EAE, nous évaluons les modalités de reprogrammation des lymphocytes T régulateurs à des fins thérapeutiques.

AXE 2: Rôle des facteurs environnementaux dans la régulation des lymphocytes T dans la sclérose en plaques.

La sclérose en plaques (SEP) est l’une des principales causes de handicap neurologique affectant plus de 2,8 millions de personnes dans le monde. Il n’existe actuellement aucun remède contre la SEP, car l’étiologie de la maladie reste pour l’essentiel inconnue. La SEP est un trouble inflammatoire et démyélinisant chronique du système nerveux central (SNC) qui résulte d’une dérégulation immunitaire, provoquée par une interaction complexe entre une prédisposition génétique et des facteurs environnementaux. Les lymphocytes T jouent un rôle clé dans la SEP, avec des fonctions régulatrices défectueuses.

L’homéostasie des lymphocytes T est contrôlée par une série de protéines exprimées à leur surface qui contrôlent leur activation et leur différenciation, notamment le régulateur du complément CD46. L’altération de la voie CD46 est décrite dans un certain nombre de maladies inflammatoires chroniques, dont la SEP, avec une induction dysfonctionnelle des cellules Tr1 sécrétant de l’IL-10.

Parmi les facteurs environnementaux liés à la SEP, la carence en vitamine D est un problème de santé publique croissant proposé comme facteur de risque majeur, avec une corrélation établie entre de faibles niveaux de vitamine D et une incidence et une gravité accrues de la SEP. Cependant, les mécanismes impliqués sont encore largement inconnus.

Notre groupe se concentre sur les mécanismes régulant l’activation des lymphocytes T humains et sur la manière dont les facteurs environnementaux, tels que la carence en vitamine D, affectent l’activation des lymphocytes T et peuvent contribuer à la pathogenèse. Nous étudions également comment les polymorphismes des gènes associés à un risque accru de SEP contribuent à cette interaction complexe.

Nos principaux objectifs sont de comprendre la régulation de la différenciation des cellules T et notamment des cellules Tr1, et de déterminer l’impact de la vitamine D dans les cellules T. Nous disséquons les mécanismes moléculaires impliqués dans l’utilisation de cellules T humaines primaires provenant de donneurs sains et de patients atteints de SEP. Notre objectif ultime est de découvrir de nouvelles voies qui pourraient fournir de nouvelles cibles immunothérapeutiques pour la SEP.

AXE 3: Rôle des molécules de signalisation des lymphocytes T dans les maladies inflammatoires du système nerveux central
Le développement d’une réponse immune adéquate et contrôlée est cruciale pour maintenir l’homéostasie de l’’organisme. Une réponse déséquilibrée peut conduire à des maladies auto-immunes si le système immunitaire réagit trop fortement ou permet une expansion tumorale due à une réponse immunitaire insuffisante. Par conséquent, la compréhension des mécanismes de contrôle des lymphocytes T est essentielle pour développer de nouvelles stratégies thérapeutiques. La signalisation après l’engagement des récepteurs exprimés par les lymphocytes T joue un rôle important dans le contrôle des fonctions des lymphocytes T. Ainsi, un défaut quantitatif ou fonctionnel ces molécules de signalisation pourrait prédisposer aux maladies autoimmunes.

À cet égard, nos études récentes ont révélé l’implication de Vav1, Themis1, CD46 et CD226 dans les fonctions des cellules T et la susceptibilité aux troubles immunitaires. Cependant, les mécanismes cellulaires et moléculaires par lesquels ces molécules de signalisation coopèrent pour réguler la fonction des cellules T restent inconnus.

Ainsi, les objectifs de notre projet sont :
1 – De déterminer, en utilisant de nouveaux modèles murins génétiquement modifiés, les mécanismes cellulaire et moléculaire par lesquels le hub de signalisation Themis1/Vav1 prévient l’apparition des troubles auto-immuns ou inflammatoires.
2 – D’analyser comment CD226 et CD46 contribuent au développement d’une réponse immune aberrante chez les patients atteins de sclérose en plaques.
3 – D’analyser comment les facteurs environnementaux, ou leurs carences telles que les rayonnements ultraviolets, affectent l’activation des lymphocytes T et peuvent contribuer à la pathogenèse

AXE 4: Etude des facteurs de transcription dans les réponses immunitaires normales et pathologiques

L’objectif principal de notre groupe consiste à déterminer le rôle des facteurs de transcription dans l’inflammation du système nerveux central

Nous avons récemment démontré que le facteur de transcription Foxo3 joue un rôle clé dans la physiopathologie de l’auto-immunité du SNC puisque la déficience en Foxo3 est associée à une diminution significative de la sévérité de l’EAE. Nous avons également mis en évidence que cette diminution résultait de l’incapacité des lymphocytes T CD4+ déficients en Foxo3 à se différencier en cellules T helper-1 (Th1) pathogènes, caractérisés par la production d’Interféron-g (IFN-g) et du facteur de stimulation des colonies de granulocytes-macrophages (GM-CSF). Au niveau moléculaire, nous avons également mis en évidence quela voie Foxo3-Eomes est essentielle pour la mise en place du programme génétique nécessaire pour la différenciation des cellules Th1 pathogènes et le développement de la neuroinflammation.

Ainsi, notre projet actuel repose sur l’analyse des fonctions du facteur de transcription Eomes en conditions normales et pathologiques. En parallèle, nous chercherons à évaluer la pertinence de nos résultats chez l’Homme en étudiant l’impact de la voie FOXO3/EOMES sur la fonction de cellules T provenant de donneurs sains ou de patients atteints de SEP.

Nous développons également un projet visant à étudier le rôle des facteurs de transcription dans la gestion de la mort neuronale dans les maladies neurodégénératives. En effet, nous avons récemment observé que des souris déficientes en Foxo3 sont résistantes à l’induction de la mort neuronale dans un modèle de maladie de Parkinson. Nous tentons aujourd’hui d’élucider la part de responsabilité des différents types cellulaires (neurones, microglie, T CD4) déficients en Foxo3 dans cette neuroprotection, de caractériser leur état traductionnel et métabolique, et étudions leurs interactions. Nous espérons ainsi proposer de nouvelles pites thérapeutiques visant à la fois la mort neuronale et la neuroinflammation dans ces pathologies multifactorielles.

Impact sociétal

Notre équipe a pour objectif de comprendre les déterminants cellulaires et moléculaires impliqués dans les maladies inflammatoires et auto-immunes qui affectent le système nerveux central. Dans cet objectif, nous avons développé des modèles expérimentaux originaux sur lesquels des recherches innovantes peuvent être menées. Nous avons également collecté des ressources biologiques uniques de patients atteints de SEP ou de narcolepsie qui nous permettent de tester la pertinence de nos principaux résultats expérimentaux chez l’Homme (validation des voies contribuant à la susceptibilité aux maladies, identification de nouveaux biomarqueurs et initiation d’essais thérapeutiques).

Notre programme scientifique s’appuie en partie sur l’analyse approfondie de voies clés que nous avons identifiées au cours des dernières années (bi-réactivité, Vav1, Themis, TYK2). En outre, nous proposons maintenant des projets émergents liés à des sujets pour lesquels nous avons réalisé des observations originales (Foxo3, Eomes, Blimp, CD226) ou générés des outils uniques (narcolepsie, maladie neurologique paranéoplasique). Notre équipe est composée de chercheurs confirmés et de jeunes chercheurs issus d’origine et de formation diverses (médecin, scientifique, vétérinaire, pharmacien …). Cela favorise un environnement scientifique dynamique et offre des opportunités uniques pour développer des recherches fondamentales et transrationnelles. Nous bénéficions d’une visibilité internationale solide dans le domaine de la neuroimmunologie. Des collaborations nationales et internationales du plus haut niveau scientifique ont été construites au cours des années. Certaines de nos recherches actuelles ont suscité l’intérêt de sociétés de biotechnologie comme GSK, Biogen et Genzyme qui fournissent à notre équipe d’autres sources possibles de financement. Pour ses différents projets, notre équipe a réussi à obtenir un certain nombre de subventions qu’elles soient locales, nationales ou internationales.
